Fractional composition of metal ions in humic gels

This thesis deals with bond strength of metal ions in humic gels. Several samples of humic acids were studied to consider influence of samples isolation process and their consequent characteristics on bond strength of metal ions in their complexes. Some functional groups was blocked by methylation at some samples. Amount of active functional groups in individual samples was determined by standard titration methods. Metal ions bound in humic complexes were divided into several fractions by extraction with different reagents (distilled water, 1M MgCl2, 1M HCl and 0,025M NH4EDTA). Two extraction methods were used: sequential and fractional. During the fractional extraction leaching of metal ions always proceeds in single agents from original komplex, whereas during the sequential extraction leaching proceeds gradually in all extraction agents in order according to rising afinity of agents, i.e. bond strength of metal ions in samples. Amount of metal ions in leaches after extractions was analysed by UV-VIS spektrometry.
Use of thin layer chromatography for fractionation of humic substances isolated from oxyhumolite

The main goal of this thesis was to optimize the fractionation of humic substances (HS) using the method of Thin-layer chromatography, so that it can be used for physico-chemical characterization of the obtained individual organic fractions. Due to the high content of HS in the natural oxyhumolite matrix this oxidized brown coal was selected as a source of these biocolloid compounds which were isolated according to the international IHSS procedure. The sample of native oxyhumolite and oxyhumolite after extraction was characterized by thermal techniques such as thermogravimetric (TGA) and elemental analysis (EA). The above-mentioned oxyhumolite samples were further analyzed by Fourier transform infrared spectroscopy, mainly for deeper understanding of their structure in terms of an organic matrix containing HS. Characterization of isolated HSs was performed by the following thermochemical and spectrometric techniques, elemental analysis, thermogravimetry, ultraviolet and visible electromagnetic absorption (UV/Vis) molecular spectroscopy, Fourier transform infrared spectroscopy and steady-state fluorescence spectroscopy. Molecular absorption spectroscopy (UV/Vis) and steady-state fluorescence spectroscopy were used for physicochemical characterization of individual organic fractions.
Experimental study on the release of active ingredients from gel matrices

This thesis deals with the characterization of hydrogels containing humic substances and salicylic acid. Experimental study of the transport of active substances was performed from the gel matrix over the skin and synthetic membranes. During this work was writing literature review focused on the use of gels with humic substances in cosmetic, medical and pharmaceutical industries. The model humic hydrogels was prepared with salicylic acid and their characterization by basic methods of material analysis. These basic methods were and visual assessment of consistency during the preparation and rheology. The study of the release of active and humic substances was performed due to the vertical diffusion cells. The amount of released humic substance was characterized by UV-VIS method and the amount of released salicylic acid from gel matrix was determined by HPLC.
Transport of humic substances into leafs

This bachelor thesis simulates leaf foliar fertilization and describes transport processes of humic substances. Experiments were performed for two areas. The first focused on the flow through the plant cuticle as the main selective barrier. In the second one the transport of humic substances to the leaf was studied. The diffusion process was evaluated by the concentration loss of lignohumate solutions, which served as the medium of humic substances in the solution - leaf model. The results showed the effect of cuticle on nutrient penetration and its dependence on its condition. New findings are the transfer processes of humic substances into the leaves, that have proved effective in a short period of time, mainly at higher concentrations.
Preparation of humic hydrogels for appliaction in cosmetics

This bachelor´s thesis is focused on the preparation and characterization of hydrogels containing humic substances and assessment of their potential usefulness in cosmetic practice. Main objective of the work was to develop a literature review focusing on the potential use of humic substances in the cosmetics and pharmaceutical industries. Furthermore, the aim was to prepare model hydrogels addition of soluble humic substances in traditional cosmetic compositions and their characterization by basic methods of analysis parameters. The main investigated parameters were consistency resulting gels (assessed subjectively and subsequently by rheology), their internal pH and dry matter. The experimental results showed that incorporation of humic substances into hydrogels resulting gels acceptable characteristics for further use in cosmetic applications.
Swelling behavior and bioactivity of humic acids hydrogels

This thesis deals with the study of the biological activity of humic substances. Based on the literature review preparation procedures were optimized in order to develop new hydrogel forms containing humic substances and biopolymers with high swelling capacity. Besides methodology for the rapid assessment of the biological activity obtained in laboratory conditions were designed and tested. The experimental part focused on characterization of hydrogel samples by means of routine physico-chemical methods, the main attention was paid to sorption of water and release of humic substances into solution. Then the prepared hydrogel materials were tested focusing on the growth effects on the maize. The absorption of mineral nutrients by the roots of plants were studied as well. The motivation of this work comes from the potential of agricultural and environmental applications of such hydrogel materials resulting in a renewal of previously exhausted organic content of soils and consequent interruption of substantial soil degradation.
Use of thin layer chromatography for fractionation and characterization of organic matter isolated from alginite

This diploma thesis is focused on the study of structure and physicochemical properties of organic fractions of humic substances, which were obtained by the method of thin-layer chromatography. Humic substances, which were used in this study, were isolated from a sample of Slovak alginite based on the procedure of the International Humic Substances Society (IHSS). The following analytical techniques were selected for the characterization of isolated humic substances: thermogravimetric and elemental analysis, molecular absorption spectroscopy (UV/Vis), Fourier transform infrared spectroscopy (FTIR) and steady-state fluorescence spectroscopy. Each organic fraction of humic substances were characterized by molecular absorption spectroscopy (UV/Vis) and steady-state fluorescence spectroscopy. Using steady-state fluorescence spectroscopy, humic substances were found to be composed of fluorophores of humic and non-humic (protein) character. Organic fractions corresponding to the humic fluorophores were characterized by a higher content of oxygen substituents on the aromatic nukleus, a higher degree of aromaticity and also a higher molecular weight. The aim of this diploma thesis was to design and optimize the process of organic matter fractionation for the purpose of detailed understanding of the structure and properties of humic substances, which were isolated from the sedimentary rock alginite. Based on this, the practical applicability of the thin layer chromatography method to significantly reduce the molecular heterogenity of the studied humic substances was evaluated.
Transport of metal ions in humic hydrogels

This diploma thesis is focused on the study of diffusion of metal ions in hydrogel of humic acid. The objective of this thesis was to investigate the effect of the concentration of metal ions supply source and duration of diffusion on transport through the gel using method of diffusion couple. Cupric, nickel and cobalt ions were esed like the diffusion medium. All these ions were selected thanks to their high affinity for the humic acids and because they are ideal to investigate the diffusion through the humic gel. Chloride salts of these metals, namely CuCl2, NiCl2 and CoCl2, were used like supply sources. Three different concentrations of each solution were compared, specifically 0,05M, 0,1M and 1M, and also were three different durations of diffusion, specifically 10 hours, 1 day and 3 days. Metal ions were compared with each other in order to determine which one has the best transport properties through the humic hydrogel. The data, necessary for calculation, were obtained by UV-VIS spectrophotometry. Diffusion coefficients and duffusive flux of individual metal ions were determined from measured data.
Production, Characterization and Design of Applications of Regenerated Humic Acids

Předkládaná disertační práce se zabývá huminovými látkami (HL), zejména huminovými kyselinami a jejich solemi, tj. humáty. V práci je prezentována literární rešerše o huminových látkách, stručně je zmíněna historie výzkumu huminových látek, obsáhle jsou pak prezentovány práce o jejich struktuře a supramolekulovém uspořádání. Jsou též zmíněny metody extrakce huminových látek z různých zdrojů. Dále jsou v tomto pojednání zařazeny a diskutovány práce zabývající se biologickými „hormonálními“ vlastnostmi HL. Z hlediska chemie životního prostředí jsou zmíněny zejména sorpční vlastnosti HL. Popsané průmyslové aplikace HL pak zahrnují celou řadu patentů a publikovaných prací, zabývající se využitím huminových látek jakožto barviv či přísad do polymerů, atd. V experimentální části je popsána příprava regenerovaných lignitů, z nichž jsou dále extrahovány huminové kyseliny a připraveny huminové soli, tzv. humáty. Princip regenerace spočívá v oxidaci původního lignitu širokou koncentrační řadou kyseliny dusičné a peroxidu vodíku. K charakterizaci získaných huminových materiálů je výhodně použit nový přístup, kdy jsou kombinovány výsledky z analýz chemických (elementární analýza, infračervená spektroskopie, termogravimetrie) a fyzikálně-chemických (dynamický rozptyl světla, spektrometrie relaxační nukleární magnetické rezonance, vysoceúčelná velikostně-vylučovací chromatografie a fluorescenční spektrometrie se zhodnocením hydratace humátů pomocí vysocerozlišovací ultrazvukové spektrometrie a hustoměru). Tyto metody jsou navíc položeny do kontextu s charakterizací biologické aktivity humátů, která je provedena pomocí modifikované metody založené na měření délky a hmotnosti kořenů kukuřice a laterálních kořenů. V závěru je použit statistický přístup s využitím Pearsonova korelačního koeficientu a jsou též (na základě výsledků pilotních studií) navrženy dvě potenciální environmentální aplikace regenerovaných huminových materiálů – sorpce antibiotika tetracyklinu a využití regenerovaného lignitu jako zdroje zkvasitelných cukrů.
Application of trihalomethane formation potential for evaluation of drinking water treatment efficiency

The main theme of this thesis is the usage of the test investigating the formation potential of trihalogenmethanes (THM FP) and its comparison with conventional indicators of water quality (CODMn, A254 and colours). As for the experimental part of the thesis, rating of the effectiveness of selected technological procedures has been applied (e.g.: coagulation, adsorption and ozonation in the act of treatment of artificially prepared water model with a higher content of humic substances in laboratory conditions). Based on the obtained results, it was found that the common indicators of water quality have very similar attributes and also complete each other. On the contrary, those attributes of indicators may not be sufficient in case of the evaluation of water quality with high content of humic substances. The findings made illustrate that the THM FP test enables the accurate evaluation of water quality, especially in connection with the content of an organic material susceptible to chemical changes. THM formation potential has proved its high value and also has confirmed that it can appropriately complete the other standarts that are being used for assessing the quality of treated water up to nowadays.
